Do the bonus round puzzles have to include a vowel? Could a puzzle like "GYPSY" be used now?
I read somewhere before that the puzzle MUST have at least one of the given letters (RSTLNE) showing but I never knew if it must have a vowel.

I don't think so. I believe that I have seen a few bonus puzzles where RSTLNE did not appear at all.

I know they've had MYTHS as a bonus puzzle sometime around 2004, so I wouldn't say that having at least one of AEIOU is a requirement — it's more like there are just so few BR-worthy answers that don't have AEIOU. GYPSY and LYNYRD SKYNYRD both seem like perfectly cromulent bonus puzzles, except for the fact that they no longer use 5-letter anwers in the BR (except for GLOBE in 2011), and they haven't used Proper Name in the BR since 2008.
The last bonus puzzle I have as not having RSTLNE at all was PIKACHU in 2001, so it's probable that the "must have at least one of RSTLNE" rule was not imposed until after then.
